A firefighter with the Prentiss County Rural Fire Department said four motorcycles, a truck and a car were involved in the collision. The truck was carrying two jet skis.
It took place at the intersection of the Highway 30 Bypass and County Road 5031 Saturday afternoon, just outside the Booneville city limits.
Two helicopters carried the two injured to North Mississippi Medical Center in Tupelo.
Neither the names of those killed nor those injured have been released by authorities pending notification of next of kin.
Prentiss County Coroner Greg Sparks said the preliminary cause of death for both individuals is multiple blunt-force trauma.
The accident is under investigation by the Mississippi Highway Patrol.
